Both cutaneous neoplasms can be a significant cause of economic losses, morbidity and mortality in the animals. The treatment of equine skin cancer can be challenging and established, evidence-based therapies resulting in sustained tumor regression are rare. The pentacyclic, lupane-type triterpenes betulinic acid (BA) and betulin can be isolated from many botanical sources, predominantly from the bark of white birch and plane trees. In addition to various biological properties, they have gained attention mainly due to their anticancer features. Within the framework of the current PhD project, the compounds BA, BA derivative NVX-207, and betulin derivative betulinyl-bis-sulfamate were assessed in various in vitro and in vivo studies for their potential as a topical therapy for ES and EMM. In conclusion, the results generated from cell culture experiments, skin permeation assays and studies in the target species are promising. Further advancement of the investigational medicinal products studied herein could lead to an effective topical and marketable, novel drug which helps to relieve suffering and, consequently, improve the welfare of equine skin cancer patients.

The aim of the Special Issue was to generate a compilation of in vitro and in vivo strategies based on cell–material interactions. Antiviral drugs have been successfully developed and used clinically for a limited number of important human viral diseases notably caused by human immunodeficiency virus (HIV), hepatitis C virus (HCV), hepatitis B virus (HBV), herpes, and influenza viruses. Despite the successes of these antiviral drugs, issues with drug resistance and toxicity remain challenging. These challenges are driving research to identify new drug candidates and to investigate novel drug targets to develop new mechanistic drug classes. Antiviral agents are not available against many viruses that cause human disease and economic burdens; in particular, the development of antiviral agents against emerging, re-emerging, and neglected viruses is increasingly becoming a priority. This book includes six review articles that discuss new antiviral strategies. The reviews either discuss advances relating to a specific virus or new therapeutic targets and approaches. Kolobov, 2023-11-09 Low-temperature plasmas (LTP) at atmospheric gas pressure play an increasing role in biomedical applications. The experimentally observed benefits of LTP for these applications are attributed to the controllable fluxes of chemically active species that can be produced in air at near room temperatures and delivered to bio-matter to induce desired effects. Recent research on the biomedical applications of LTP has generated new scientific knowledge regarding the interaction of plasma with soft matter including cells, tissues, seeds, and plants. The observed effects of LTP on biological cells are mediated by the plasma-produced reactive oxygen species (ROS) and reactive nitrogen species (RNS). These include hydroxyl, OH, atomic oxygen, O, singlet delta oxygen, O2(1Δ), superoxide, O2-, hydrogen peroxide, H2O2, and nitric oxide, NO. Some of these species are known to play important roles in biology serving as signaling molecules in living organisms. When they come in contact with biological cells these species interact with the lipids and proteins of the cell membrane, enter the cell and increase the intracellular ROS concentrations, which can lead to DNA damage and may compromise the integrity of other cell organelles. ROS and RNS can also trigger cell signaling pathways, which can lead to cellular death by apoptosis or necrosis. Other plasma-generated agents that could play biological roles are charged particles (electrons and ions), UV photons, and electric fields.

Biomaterials like polyglycolic acid, collagen and small-intestinal submuscosa provide a temporary scaffold to guide new tissue growth and or- nization. Typically, they need to be biodegradable, showing good cell atta- ment and proliferation and they should possess appropriate mechanical properties (Kim et al. , 2000). Synthetic polymers ful ll most of these requirements but lack cell-adhesion peptides on their surface to enhance cell attachment. Ce- adhesion peptides are present in ECM proteins like collagen and elastin. Thus a synthetic polymer coated with ECM proteins would result in a scaffold that mimics the natural cellular environment with enhanced cell attachment and p- liferation. The new bioactive scaffold will be made by combining a synthetic polymer coated with a layer of recombinant ECM proteins produced by CHO cells. The rst step consists of identifying polymers that give best results in terms of CHO cell attachment and growth. Classical techniques to determine biomass are inappropriate to evaluate 3-D structures. Thus a screening system based on stable GFP expressing CHO cells was used to compare the different scaffolds. Simple uorescent measurement after cell lysis allows determining cell attachment and p- liferation on synthetic polymers. An additional allotropic modification of oxygen, the cyclooctaoxygen, was predicted to exist in 1990. The first synthesis and characterization of cyclooctaoxygen as its sodium crown complex, isolated in the form of three cytosine nucleoside hydrochloride complexes, was reported in 2016. Cyclooctaoxygen sodium was synthesized in vitro from atmospheric oxygen, or catalase effect-generated oxygen, under catalysis of cytosine nucleosides and either ninhydrin or eukaryotic low-molecular weight RNA. Thin-layer chromatographic mobility shift assays were applied on specific nucleic acids and the cyclooctaoxygen sodium complex. The cationic cyclooctaoxygen sodium complex was shown to bind to nucleic acids (RNA and DNA), to associate with single-stranded DNA and spermine phosphate, and to be essentially non-toxic to cultured mammalian cells at 0.1–1.0 mM concentration. It is postulated that cyclooctaoxygen is formed in most eukaryotic cells in vivo from dihydrogen peroxide in a catalase reaction catalysed by cytidine and RNA. A molecular biological model was deduced for a first epigenetic shell of eukaryotic in vivo DNA. This model incorporates an epigenetic explanation for the interactions of the essential micronutrient selenium (as selenite) with eukaryotic in vivo DNA. The sperminium hydrogen phosphate/cyclooctaoxygen sodium complex is calculated to cover the actively transcribed regions (2.6%) of bovine lymphocyte interphase genome. Cyclooctaoxygen seems to be naturally absent in hypoxia-induced highly condensed chromatin, taken as a model for eukaryotic metaphase/anaphase/early telophase mitotic chromatin. Hence, it is proposed that the cyclooctaoxygen sodium-bridged sperminium hydrogen phosphate and selenite coverage serves as an epigenetic shell of actively transcribed gene regions in eukaryotic 'open' euchromatin DNA. It is proposed that the sperminium phosphate/cyclooctaoxygen sodium complex coverage of nucleic acids is essential to eukaryotic gene regulation and promoted proto-eukaryotic evolution. Cyclooctaoxygen sodium-bridged sperminium hydrogen selenite is calculated to serve as a marker shell component at ATG start codons in human euchromatin DNA mRNA genes, both at the translation initiation triplet and at 5′-untranslated region upstream ATGs. The total herbicide glyphosate (ROUNDUP®) and its metabolite (aminomethyl)phosphonic acid (AMPA) are proved to represent 'epigenetic poisons', since they both selectively destroy the cyclooctaoxygen sodium complex.
